The pCITE®-2a-c(+) vectors are designed for enhanced translation of cloned sequences in vitro using mammalian cell extracts, e.g. rabbit reticulocyte lysate. The maps for pCITE-2b(+) and pCITE-2c(+) are the same as pCITE-2a(+) with the following exceptions: pCITE-2b(+) is a 3793 bp plasmid; it lacks Nde I, EcoR V, BstX I and Xcm I sites. Subtract 7 bp from all sites beyond Msc I at 522. pCITE-2c(+) is a 3795 bp plasmid; it lacks Nde I, EcoR V, BstX I and Xcm I sites. Subtract 5 bp from all sites beyond Msc I at 522. The f1 origin in pCITE-2a-c(+) vectors is oriented so that infection with helper phage wil produce virions containing single stranded DNA that is complementary to the strand shown on the vector map. Therefore, single stranded sequencing should be performed using the CITE primer.
